The Dutch was too strong in the bantamweight for the American Julianna Peña.
De Randamie made the difference in Abu Dhabi in the third round.
The 36-year-old from Utrecht got Peña in a stranglehold, as a result of which the American five years younger could no longer breathe and the worst of it.
Immediately after her victory, De Randamie expressed her hope for a title fight with Amanda Nunes.
The two already competed for the world title in the bantamweight class in December and then the Brazilian MMA fighter won after a jury decision.
The fight against Peña was De Randamie's first appearance since the showdown with Nunes in December.
Of her last seven fights, 'The Iron Lady' won six.
De Randamie, who competes in both kickboxing and Mixed Martial Arts (MMA), became the first featherweight world champion in February 2017.
That title was taken from her in June 2017, because De Randamie refused to fight Cristiane Justino because of the Brazilian's doping past.
Since July, the UFC has been organizing fights on 'Fight Island' in Abu Dhabi, where a bubble has been organized.
That way, fighters from all over the world can take action during the corona crisis.
