Matsuyama, who took the lead the day before in the second round, returned for the second consecutive day a card of 68, two strokes under par.
Despite a bogey on the 18th and last hole of the day, he has a total of 200 (-10) ahead of American Cameron Tringale (201, -9).
Matsuyama, 19th in the world and winner of the Augusta Masters last April, covets the seventh title of his career on the PGA Tour.
A trio made up of Colombian Juan Sebastián Munoz, American Brendan Steele and Englishman Matt Wallace share third place, but are four strokes behind the leader (204, -6).
The American Collin Morikawa, 3rd in the world, is in 8th place, six lengths behind the leader, but he has just chained two cards of 68 and 67 which allowed him to get closer to the lead.
